General and Regional Circulatory Effects of Synthetic Bradykinin in Man.

چکیده
• Bradykinin is a potent vasodilator in several animal species. The circulatory effects of bradykinin in man have been less extensively studied. Fox et al. observed that intra-arterial or intravenous administration of bradykinin increased blood flow to the hand and forearm. These investigators also found that blood flow to the skin of the hand, forearm, face, and neck increased during the administration of bradykinin. Burch and DePasquale* reported that intra-arterial injection of bradykinin produced digital vascular responses similar to those seen during reactive hyperemia. They concluded that bradykinin selectively closed digital arteriovenous anastomoses. Gersmeyer and Spitzbarth found that single intravenous injections of 50 to 100 fig of bradykinin caused reduction in arterial blood pressure, acceleration of heart rate, increase in cardiac output and stroke volume, and fall in systemic vascular resistance. Comparable doses injected into the superior vena cava or pulmonary artery increased pulmonary arterial pressure. Injections of bradykinin into the femoral artery caused increased blood flow to the calf and rise in skin temperature. Saameli and Eskes •"' reported that intravenous infusions of 0.25 to 0.5 ftg of bradykinin per minute per kilogram body weight lowered arterial blood pressure and accelerated heart rate.

• It is now well established that the endogenous nonapeptide bradykinin has important effects on the circulatory system" and, in addition, may play a role in many physiologic and pathologic clinical conditions." Since its discovery in 1948,' it has been shown in animals and man that bradykinin reduces systemic arterial pressure," increases limb blood flow," and elevates cardiac output.' "• Litt...
متن کاملDigital vascular responses to intra-arterial injections of bradykinin, kallidin, and eledoisin in man.
DOISIN are vasoactive polypeptides recently synthesized in pure form.1-3 Bradykinin and kallidin exist in man, whereas eledoisin has been found only in the posterior salivary glands of Eledone. Although the three poly-peptides have potent vasodilating and hypo-tensive properties, only bradykinin and kallid-in are closely related structurally (fig. 1). The nonapeptide structure of bradykinin was...
متن کاملPreparation of a Photoactivable Ligand for Bradykinin Receptors
ABSTRACT Most of physiological effects of bradykinin is due to its effect on bradykinin B2 receptors. There is no reports of preparation of a photoactivatable analogue of bradykinin reactive with sloubilized bradykinin B2 recpetors. Photoactivatable radioactive ligands are powerful tools in purification of receptors. In this study we report preparation of a photoactivatable iodinatable analo...
متن کاملNoscapine antagonizes vasoconstrictor action of bradykinin in isolated human umbilical artery
Abstract Background: It has been demonstrated that noscapine, an antitussive opioid alkaloid, could antagonize bradykinin- induced responses such as bradykinin effects in guinea-pig ileum, cough induced by bradykinin receptor agonist and angiotensin converting enzyme inhibitors, and brain damage after brain edema both in neonatal rat model and in patients with stroke. In the present study, the...
متن کاملHypoxic-ischemic encephalopathy in a young man due to tramadol overdose
Objective: Tramadol is a synthetic analgesic with two mechanisms. The opioid and non-opioid mechanisms are responsible for tramadol side effects. Non-opioid side effects of tramadol are due to the reuptake inhibitions of serotonin and norepinephrine. Some of the side effects include anaphylactoid reactions, CNS depression, hypoglycemia, hypotension, respiratory depression, seizures, and seroton...
